8月2日,華中農(nóng)業(yè)大學(xué)匡漢暉教授研究團(tuán)隊(duì)以“Loss-of-function of SAWTOOTH 1 affects leaf dorsiventrality genes to promote leafy heads in lettuce”為題在The Plant Cell上發(fā)表了研究論文。該研究克隆了控制生菜結(jié)球的新基因LsSAW1,發(fā)現(xiàn)其可以下調(diào)近軸基因和上調(diào)遠(yuǎn)軸基因以促進(jìn)生菜結(jié)球發(fā)育。
課題組前期已克隆了首個(gè)控制生菜結(jié)球的基因LsKN1(Lactuca sativa KNOTTED 1),其上調(diào)表達(dá)會(huì)促進(jìn)葉球發(fā)育,但僅有LsKN1是不夠的。在前期研究基礎(chǔ)上,作者成功構(gòu)建了一個(gè)結(jié)球與不結(jié)球表型分離的F5群體,且在該群體中,結(jié)球是隱性性狀,結(jié)球個(gè)體近軸部分的細(xì)胞排列紊亂。利用BSA與RNA-seq分析,作者發(fā)現(xiàn)該群體中結(jié)球性狀是由LHL1位點(diǎn)獨(dú)立控制的,經(jīng)過圖位克隆與序列分析,作者發(fā)現(xiàn)LHL1位點(diǎn)中LG4_402589外顯子上存在一個(gè)堿基的缺失,導(dǎo)致無法翻譯出完整的蛋白,該基因與擬南芥SAW1同源,命名為L(zhǎng)sSAW1。經(jīng)過反向遺傳學(xué)驗(yàn)證,作者證實(shí)了LHL1位點(diǎn)就是LsSAW1,其功能缺失(Lssaw1)對(duì)生菜結(jié)球的發(fā)育起到關(guān)鍵作用。
關(guān)于LsSAW1如何調(diào)控生菜結(jié)球發(fā)育,作者通過原位雜交發(fā)現(xiàn)LsSAW1主要在不結(jié)球植株葉片的近軸部表達(dá),但在結(jié)球植株葉片的任何部位都檢測(cè)不到它的表達(dá),作者推測(cè)這種低表達(dá)可能由啟動(dòng)子的SNP引起,也可能是由外顯子的突變引起。通過RNA-seq作者發(fā)現(xiàn)參與葉背腹極性有關(guān),于是作者進(jìn)一步分離了葉片近軸與遠(yuǎn)軸細(xì)胞進(jìn)行RNA-seq,發(fā)現(xiàn)生長(zhǎng)素信號(hào)與生長(zhǎng)控制相關(guān)基因在在Lssaw1植株的DEGs中顯著富集。進(jìn)一步分析近軸與遠(yuǎn)軸部的差異基因發(fā)現(xiàn),Lssaw1下調(diào)了近軸基因,上調(diào)了遠(yuǎn)軸基因,從而促進(jìn)了生菜葉球的發(fā)育。
為了證實(shí)Lssaw1通過調(diào)節(jié)近軸與遠(yuǎn)軸基因促進(jìn)葉球發(fā)育的假說,作者選擇了一個(gè)近軸基因LsAS1進(jìn)行進(jìn)一步分析,發(fā)現(xiàn)LsAS1主要在不結(jié)球植株葉片的近軸部表達(dá),而Lssaw1突變體中表達(dá)量顯著下降。通過CHIP-seq,酵母單雜以及EMSA實(shí)驗(yàn)均證實(shí)了LsSAW1與LsAS1的啟動(dòng)子結(jié)合,Lssaw1下調(diào)LaAS1的表達(dá),遺傳實(shí)驗(yàn)也表明LaAS1下調(diào)會(huì)促進(jìn)葉球發(fā)育,上調(diào)則會(huì)抑制葉球發(fā)育。作者對(duì)遠(yuǎn)軸面基因LsYAB1進(jìn)行類似的實(shí)驗(yàn)發(fā)現(xiàn),在Lssaw1突變體中LsYAB1的抑制作用減弱,有助于葉球發(fā)育。
作者希望通過酵母篩庫來找到與LsSAW1一起調(diào)控葉片背腹性的蛋白,結(jié)果發(fā)現(xiàn)LsSAW1與之前研究的LsKN1存在互作,進(jìn)一步研究發(fā)現(xiàn),LsSAW1與LsKN1在葉球發(fā)育中的作用是相互拮抗的。例如LsSAW1可以直接與LsAS1啟動(dòng)子結(jié)合并上調(diào)其表達(dá),同時(shí)減弱LsKN1對(duì)LsAS1的抑制作用,再次證實(shí)了Lssaw1可以直接和間接下調(diào)LsAS1的表達(dá)促進(jìn)葉球發(fā)育。此外LsSAW1的同源基因LsSAW2在Lssaw1背景下過表達(dá)同樣可以導(dǎo)致生菜不結(jié)球,但是一般情況下由于LsSAW2表達(dá)水平過低,導(dǎo)致其無法發(fā)揮冗余作用。
作者對(duì)488份生菜野生祖先進(jìn)行重測(cè)序發(fā)現(xiàn),竟然沒有一份材料在Lssaw1中有一個(gè)堿基的缺失,但在21個(gè)現(xiàn)代品種中出現(xiàn)了這種突變,表明Lssaw1突變是人類馴化(好吃)的結(jié)果。體現(xiàn)出該突變對(duì)生菜育種以及葉球發(fā)育的重要性。
華中農(nóng)業(yè)大學(xué)安光輝博士為論文第一作者,加州大學(xué)戴維斯分校Dean Lavelle與Richard W. Michelmore也參與該項(xiàng)研究,Elizabeth Georgian與康春穎教授對(duì)文章提出寶貴建議。本研究得到國家自然科學(xué)基金的資助。

The mechanisms underlying leafy heads in vegetables are poorly understood. Here, we cloned a quantitative trait locus (QTL) controlling leafy heads in lettuce (Lactuca sativa)。 The QTL encodes a transcription factor, SAWTOOTH 1 (LsSAW1), which has a BEL1-like homeodomain and is a homolog of Arabidopsis thaliana. A 1-bp deletion in Lssaw1 contributes to the development of leafy heads. Laser-capture microdissection and RNA-sequencing showed that LsSAW1 regulates leaf dorsiventrality and loss-of-function of Lssaw1 downregulates the expression of many adaxial genes but upregulates abaxial genes. LsSAW1 binds to the promoter region of the adaxial gene ASYMMETRIC LEAVES 1 (LsAS1) to upregulate its expression. Overexpression of LsAS1 compromised the effects of Lssaw1 on heading. LsSAW1 also binds to the promoter region of the abaxial gene YABBY 1 (LsYAB1), but downregulates its expression. Overexpression of LsYAB1 led to bending leaves in LsSAW1 genotypes. LsSAW1 directly interacts with KNOTTED 1 (LsKN1), which is necessary for leafy heads in lettuce. RNA-seq data showed that LsSAW1 and LsKN1 exert antagonistic effects on the expression of thousands of genes. LsSAW1 compromises the ability of LsKN1 to repress LsAS1. Our results suggest that downregulation or loss-of-function of adaxial genes and upregulation of abaxial genes allow for the development of leafy heads.
